seed out at ca. 0.5 x 10⁶ cells/ml; maintain at ca. 0.3 - 1.0 x 10⁶ cells/ml; split 1:2 every 2-3 days; cells grow in clumps and are difficult to disperse; viability may decrease after thawing and when dispersed too vigorously; cell counting may be difficult, better after digestion with accutase (sedimented cells, 3-5 ml accutase, 10 min)

at 37 °C with 5% CO₂
