established from the tumor of a 60-year-old man with moderately differentiated squamous cell carcinoma of the soft palate which was invasive into muscle layer; cell line was established from a cervical lymph node metastasis which occurred seven years after initial treatment of the primary tumor; the tumor metastasized not only to the lungs, but also to the brain; described as anchorage-independent and tumorigenic in nude mice

epithelial-like polygonal or round and flat cells (sometimes spindle-form, very heterogeneous) growing in multilayers

90% DMEM (4.5 g/L glucose) + 10% h.i. FBS
Subculture:
split confluent culture 1:5 every 5 days using trypsin/EDTA; seed out at ca. 1.5 x 106 cells/80 cm2 in 15 ml medium
Incubation:
at 37 °C with 5% CO2

ca. 50 hours
Harvest:
cell harvest of ca. 8-10 x 106 cells/175 cm2
