Origin:
Genetical modification of U2OS#18 (DSMZ ACC 832), a reporter cell line for studying sister chromatid recombination, SCR). This cell line belongs to the series of genetically disrupted RAD51 paralogs with reversion of the disruption by retroviral transfection with the functional gene (the non-complemented sister cell line is ACC 834). Cells are described in the literature to be puromycin and hygromycin resistant

Medium:
90% DMEM + 10% h.i. FBS + (additionally) 2 mM L-glutamine (to verify antibiotic resistance, 1 µg/ml puromycin and/or 150 µg/ml hygromycin should be added)
Subculture:
seed out at 1 x 106 cells/175 cm2;
split confluent culture 1:4 to 1:10 every 4-10 days using trypsin/EDTA
Incubation:
at 37 °C with 10% CO2
Doubling time:
ca. 25-40 hours
Harvest:
cell harvest of 15-25 x 106 cells/175 cm2 flask
