Origin:
established from the bone marrow of a woman with multiple myeloma in 1967; cells were described in the literature to synthesize IgG and to have receptors for insulin and calcitonin; cells express BCL2 mRNA; cells appear to be rather "EBV+ B-lymphoblastoid cells" than myeloma-derived cells (see also Pellat-Deceunynk et al., Blood 86: 4001-4002, 1995), hence NOT a malignant myeloma cell line

Risk assessment:
The cell line is positive for EBV (HHV-4) by PCR analysis. Additionally, expression of immediate-early protein BZLF-1 was detected by western blot after stimulating the cells with phorbol ester / sodium butyrate. The infection was classified as potentially lytic associated with a possible production of active viruses. A transmission of EBV during handling of the cells is possible and the cell line is thus categorized biosafety level 2.

Medium:
90% RPMI 1640 + 10% h.i. FBS
Subculture:
seed out at ca. 1 x 106 cells/ml; maintain at 1-2 x 106 cells/ml, split 1:2 to 1:3 every 2-3 days; as the medium becomes quickly acidic (yellow) with a corresponding cell growth, it is advisable to change the supernatant medium every 1-2 days
Incubation:
at 37 °C with 5% CO2
Doubling time:
about 45 hours
Harvest:
maximal cell density of 2-2.5 x 106 cells/ml
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
