Origin:
established from the supraorbital lymph node metastasis of a neuroblastoma of a 14-year-old girl in 1971 (see ATCC HTB 10); although described in the original paper as neuroblastoma, SK-N-MC is now widely regarded as having originated from the morphologically similar Askin's tumor related to Ewing's sarcoma; FLI1-EWS gene fusion, the molecular pathological change in Ewing's sarcoma and Askin's tumor, was shown in SK-N-MC by Dunn et al.

Subculture:
split confluent culture 1:3 to 1:6 every 3-5 days using trypsin/EDTA; seed out at ca. 1-4 x 106 cells/80 cm2; cells may grow slowly initially
Incubation:
at 37 °C with 10% CO2
Doubling time:
ca. 48 hours
Harvest:
cell harvest of about 25 x 106 cells/80 cm2
