growing mainly as adherent epithelioid cells forming a monolayer and to a lower extend as singly round cells in suspension;

Medium:
90% RPMI 1640 + 10% h.i. FBS
Subculture:
seed out at ca. 1-2 x 106 cells in a 80 cm2 flask; cells grow adherently until confluence as well as in suspension; split confluent culture every 2-4 days; if only suspension cells are used for passaging, a new monolayer will develop; adherent cells should be detached using trypsin/EDTA
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 24-30 hours
Harvest:
maximal density at ca. 1 x 106 cells/ml (cells in suspension) and 6-8 x 106 cells in 80 cm2 flasks
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma:
initial contamination was eliminated with Mycoplasma Removal Agent, then negative in DAPI, microbiological culture, RNA hybridization, and continuously in PCR assays
