Origin:
established from the peripheral blood of a 77-year-old man with acute myeloid leukemia (AML M4) in relapse in 1990; the cells are described in the literature to respond to G-CSF, GM-CSF, IL-3, and FLT3 ligand; we have noted that the cell line can become very quickly cytokine-independent (within 3-4 weeks), presumably due to rapid outgrowth of independent cells

Medium:
70% alpha-MEM (with ribo- and deoxyribonucleosides) + 20% h.i. FBS + 10% vol conditioned medium of cell line 5637 (DSM ACC 35) (or 10 ng/ml GM-CSF)
Subculture:
seed out at about 0.5 x 106 cells/ml (we recommend to use 12- or 24-well plates); maintain at 0.5-1.5 x 106 cells/ml; optimal split ratio of 1:2 to 1:4 every 2-4 days; viability may decrease after thawing, but cells recover quickly and viability will raise over the next few passages. CAVEAT: initial cytokine-dependent cells lines may become independent due to outgrowth of subclones; this selective outgrowth may be promoted by
cytokine insufficiency and inadequate cell density or long-term cell culture. Thus the DSMZ cannot guarantee indefinite stability of factor-dependence of the cell line.
