established from the poorly differentiated squamous cell carcinoma resected from upper (cervical) esophagus of a 49-year-old woman after receiving radiotherapy (tumor was invading contiguous structures); described as carrying amplified oncogenes, c-erb-B (8x) and cyclin D1 (4x) and producing tumors in nude mice; extensively characterized cell line (see cellosaurus), listed in CCLE (ref 18232)

49% RPMI 1640 + 49% Ham's F12 + 2% h.i. FBS
Subculture:
seed out at ca. 1 x 106 cells/175 cm2; split confluent culture up to 1:20 every 3-4 days using trypsin/EDTA; cells grow very fast, a FBS amount around 2-5% is sufficient
Incubation:
at 37 °C with 5% CO2
Doubling time:
below 25 hours (even less when serum amount is increased)
Harvest:
cell harvest of ca. 15 x 106 cells in a 175cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO

Mycoplasma:
initial contamination was eliminated with BM-Cyclin (tiamulin & minocycline), then negative in DAPI, microbiological culture, RNA hybridization and continuously in PCR assays
