At the DSMZ, the cyanobacterial cultures are usually cultivated without agitation in 25 cm2 plastic tissue culture flasks with filter screw caps. To subculture Coleofasciculus strains, scrape off the adhering cell material with a plastic Pasteur pipette and crush it by pipetting up and down several times. Alternatively, use a disposable pipette tip to dissect the cell material. Use 2-3 ml of the shredded biomass to inoculate 15 ml of fresh medium in a new tissue culture flask. Light cycle: 16 hours illumination at 300-350 lux and 8 hours dark period. Transfer interval of 12 weeks.
