established from the pleural effusion of a 69-year-old woman with metastatic invasive breast adenocarcinoma (after radio- and hormone therapy) in 1970; widely used and extensively characterized cell line; described as ER-positive, but HER2neu negative; listed in the NCI cancer cell line panel and in the CCLE (ref 18232); a derivative expressing GFP is available (ACC 983);

80-90% RPMI 1640 + 10-20% h.i. FBS + MEM non-essential amino acids + 1 mM sodium pyruvate + 10 µg/ml human insulin
Subculture:
seed out at ca. 1 x 106 cells/80 cm2 flask; split confluent culture 1:2 to 1:4 every 2-6 days using trypsin/EDTA; cells may grow slowly after thawing
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 50 hours (range 30-72 hours)
Harvest:
about 40-50 x 106 cells/175 cm2
