this cell line (full name: IPLB-SF-21-AE) was derived from immature ovaries of fall armyworm Spodoptera frugiperda pupae (species Spodoptera frugiperda, family Noctuidae, order Lepidoptera); cells are susceptible to Baculovirus infection; see also SF-9 (DSMZ ACC 125) which was derived from SF-21

80-90% TC 100 medium + 10-20% h.i. FBS (or 80-90% Grace´s insect medium + 10-20% h.i. FBS supplemented with L-Glutamine, hydrolyzed lactalbumin and yeastolate)
Subculture:
split culture 1:3 to 1:5 every 4-5 days; cells grow partly adherent, but the use of trypsin is not necessary, detach cells with rubber scraper; seed out at ca. 1 x 106 cells/25 cm2
Incubation:
25-29 °C without CO2 ; temperature below 23 °C may lead to reduced growth and loss of adherence
Doubling time:
ca. 3 days
Harvest:
about 25 x 106 cells/80 cm2
