SF-21 - Discontinued
ACC 119
Cell line:
SF-21 - Discontinued
DSMZ no.:
ACC 119
Species:
insect - fall armyworm (Spodoptera frugiperda) (Spodoptera frugiperda)
Cell type:
ovary cells
Origin:
this cell line (full name: IPLB-SF-21-AE) was derived from immature ovaries of the fall armyworm Spodoptera frugiperda (family Noctuidae, order Lepidoptera); susceptible to Baculovirus infection; the cell line carries the genome of the Spodoptera frugiperda rhabdovirus SfV; it is the parental cell line to cell line SF-9 (DSMZ ACC 125); the line was discontinued because it was determined by targeted sequencing of the rhabdovirus genome that it is in fact cell line SF-9
Biosafety level:
1
Permissions and restrictions:
DSMZ Cell Culture Data:
Morphology:
Medium:
80-90% TC 100 medium + 10-20% h.i. FBS (or 80-90% Grace´s insect medium + 10-20% h.i. FBS supplemented with L-Glutamine, hydrolyzed lactalbumin and yeastolate)
Subculture:
split culture 1:3 to 1:5 every 4-5 days; cells grow partly adherent, but the use of trypsin is not necessary, detach cells with rubber scraper; seed out at ca. 1 x 106 cells/25 cm2
Incubation:
25-29 °C without CO2 ; temperature below 23 °C may lead to reduced growth and loss of adherence
Doubling time:
ca. 3 days
Harvest:
about 25 x 106 cells/80 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
DSMZ Scientific Data:
Mycoplasma:
negative in DAPI, microbiological culture, RNA hybridization, PCR assays
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Spodoptera frugiperda species.
