80-90% TC 100 medium + 10-20% h.i. FBS (or 80-90% Grace´s insect medium + 10-20% h.i. FBS supplemented with L-Glutamine, hydrolyzed lactalbumin and yeastolate)
Subculture:
split culture 1:2 to 1:3 every 3-5 days (cells grow adherent, but trypsin is not necessary as cells can be detached by tapping the flask or with a rubber policeman); seed out at ca. 2 x 106 cells/80 cm2; immediately after thawing, cell viability may be low
Incubation:
25-29 °C without CO2 ; temperature below 23 °C may lead to reduced growth and loss of adherence
