90% alpha-MEM (with ribo- and deoxyribonucleosides) + 10% h.i. FBS + 2 mM L-glutamine (in case other media, e.g. DMEM or Iscove's MDM, are used, hypoxanthine and thymidine should be added as supplements)
Subculture:
seed out at ca. 0.5-2.0 x 106 cells/80 cm2; split confluent cultures 1:3 or higher every 2-4 days using trypsin/EDTA
Incubation:
at 37 °C with 10% CO2
Doubling time:
ca. 20-24 hours
Harvest:
about 10-20 x 106 cells/80 cm2 or 40-70 x 106 cells/175 cm2
