SCHNEIDER-2
ACC 130
Cell line:
SCHNEIDER-2
DSMZ no.:
ACC 130
Species:
insect - fruitfly (Drosophila melanogaster) (Drosophila melanogaster)
Cell type:
Origin:
established from the late embryo of a Drosophila melanogaster (fruitfly) in 1972; originally the cells were diploid with 5-10% XY, currently the cell line has only XX cells that are now 60-80% tetraploid; also often termed (incorrectly) S2
Reference(s):
Biosafety level:
1
Permissions and restrictions:
DSMZ Cell Culture Data:
Morphology:
Medium:
mix of Schneider's Drosophila medium + TC-100 medium (1:1) + 10-20% h.i. FBS
Subculture:
maintain at 24-30°C with gas exchange; seed out at ca. 1-2 x 106 in T25 flask; after thawing cells may have a long lag period (3-7 days) before they start to grow; split confluent culture 1:2 to 1:4 every 2-4 days; cells are resistant to trypsination, for propagation use cells in suspension or remove adherent cells with a rubber policeman
Incubation:
at room temperature up to 30 °C in normnal atmosphere
Doubling time:
ca. 40-60 hours
Harvest:
about 15-20 x 106 cells in a 25 cm2 flask
Storage:
frozen with 35% Schneider´s Drosophila medium, 35% TC-100 medium, 20% FBS, 10% DMSO
DSMZ Scientific Data:
Mycoplasma:
negative in DAPI, microbiological culture, RNA hybridization, PCR assays
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Drosophila melanogaster
Viruses:
PCR: SMRV -
