established from a neuroblastoma metastasis at an adrenal site of a 4-year-old white boy in 1986; cells were described in the literature to express neuron-specific enolase and synaptophysin (but neither GFAP nor S-100 protein) and to carry a 20-fold amplification of MYCN (NMYC, N-myc)

80-90% RPMI 1640 + 10-20% h.i. FBS + 2 mM L-glutamine + MEM non-essential amino acids
Subculture:
seed out at 1.5-3 x 106 cells/80 cm2; split confluent culture 1:2 to 1:4 every 4-6 days using trypsin/EDTA; a significant number of cells may detach spontaneously from the plastic surface and these floating cells may also be used for passaging. Note: medium must be changed frequently to prevent acidity which induces cell clumping; optimal dissociation after trypsination is affected by gentle aspiration with a #21 gauge needle and syringe

at 37 °C with 5% CO2

cell harvest of about 10 x 106 cells/80 cm2

human bimodal hyper-, diploid/tetraploid karyotype - 54(48-54)<2n>XY, +1, +1, +2, +6, +7, +8, +13, +20, der(1)t(1;?)(p32;?)x2, der(13)t(13;?2HSR)(q33;?2p23) - both the long HSR on ch 13 due to MYCN (NMYC, N-myc) amplification and the ch 1 short-arm rearrangements are typical of neuroblastoma - identity confirmed
