Medium:
80-90% RPMI 1640 + 10-20% h.i. FBS
Subculture:
seed out at ca. 0.5 x 106 cells/ml (20% FBS and a 12- or 24-well plate could be of advantage); maintain at 0.2-1.0 x 106 cells/ml; split 1:2 to 1:3 every 3-4 days; note that after thawing cell viability may drop to about 40%, but cells will recover within a week
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 40-50 hours
Harvest:
saturation density at about 1.0 x 106 cells/ml
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma:
initial contamination was eliminated with Ciprobay (ciprofloxacin), thereafter negative in DAPI, microbiological culture and repeatedly in PCR assays
