Origin:

90% RPMI 1640 + 10% h.i. FBS + 2 mM L-glutamine
Subculture:
seed out at about 0.2-0.5 x 106 cells/ml; optimal split ratio of about 1:2 to 1:3 every 2-3 days; maintain at 0.5-1.5 x 106 cells/ml
Incubation:
at 37 °C with 5% CO2
Doubling time:
about 25-35 hours
Harvest:
maximal density at about 1.5 x 106 cells/ml
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
Mycoplasma:
initial contamination was eliminated with Mycoplasma Removal Agent, then negative in DAPI, microbiological culture, RNA hybridization, and continuously in PCR assays
