90% RPMI 1640 (or mixture of RPMI 1640 + Ham's F12 at 1:1) + 10% h.i. FBS

seed out at ca. 0.5-3.0 x 10⁶ cells/175 cm²; split confluent culture 1:5 to 1:20 every 4-8 days using trypsin/EDTA
