Origin:
GOS-3 is classified as misidentified cell line with no authentic stock known (registration ID at ICLAC: ICLAC_00449). STR analysis
demonstrated that GOS-3 is genetically identical to the U-343-MG cell line. U-343-MG
was described to be established from a temporal astrocytic glioma (grade III) of a 64-year old male. GOS-3 was intended to be established from a mixed astro-oligodendroglioma (grade II/III). Since the original cell line U-343-MG is not deposited in a non-profit public collection, the misidentified GOS-3 is maintained at DSMZ. The GOS-3 cell line is part of the Cancer Cell Line Encyclopedia (CCLE) data set and is described to express neuroepithelial markers GFAP, vimentin and S-100 protein as well as pp60C-SRC, but not the neuro-specific variant pp60C-SRCN and to express neuropoietic factors CNTF, LIF and their receptors; LIF and oncostatin M induce growth inhibition and differentiation

Subculture:
split confluent culture 1:2 to 1:3 every third day using trypsin/EDTA; maintain at 7-10 x 106 cells/175 cm2; seed out at ca. 1 x 106 cells/25 cm2 in 10 ml medium

Authenticity:
STR analysis according to the global standard ANSI/ATCC ASN-0002.1-2021 (2021) resulted in an authentic STR profile of cell line U-343-MG in the reference STR database.
Cytogenetics:
the data obtained at the DSMZ are different to the published data for U-343-MG: human hypodiploid karyotype with 20% polypoidy - 44(40-45)<2n>XXY/XXYY, -6, +7, -10, -14, -22, der(1)del(1)(p21)t(1;9)(q42;q34), der(9)t(9;16)(p13;p11)t(1;9)(q42;q34), der(16)t(9;16)(p13;p11), der(18)t(6;18)(p12;q12.2) - gain of 7 and loss of 10 and 22 typical in low-grade astrocytoma-oligodendroglioma - gains of sex chromosomes (particularly Y) are very rare in tumor cells
