Medium:

90% DMEM + 10% h.i. FBS + 2 mM L-glutamine + 1 mM sodium pyruvate

Subculture:

seed out at ca. 0.5-1.0 x 10⁶ cells/175cm²; split confluent culture 1:2 to 1:4 every 3-4 days using trypsin/EDTA (incubation for about 10 min); a significant amount of cell debris is always visible in the culture background which might be due to granula or other intracellular material

Incubation:

at 37 °C with 5% CO₂

Doubling time:

ca. 70 hours

Harvest:

cell harvest of 10-15 x 10⁶ cells/175cm²

Storage:

frozen with 70% medium, 20% FBS, 10% DMSO
