80-90% DMEM + 10-20% h.i. FBS + 2 mM L-glutamine + EGF (10 ng/ml, at least in the first week)
Subculture:
seed out with 15 ml in a T75 flask; split confluent culture 1:4 to 1:8 every 3-5 days by agitation; cell number may be difficult to ascertain due to growth in clumps; with EGF cells may grow faster and in suspension
Incubation:
at 37 °C with 10% CO2
Doubling time:
ca. 40 hours
Harvest:
cell harvest of ca. 3 x 106 cells/ml or 1 x106 cells/cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
