GH4-C1
ACC 482
Cell line:
GH4-C1
DSMZ no.:
ACC 482
Species:
rat (Rattus norvegicus)
Cell type:
pituitary tumor
Origin:
subclone of the cell line GH3 (ACC 469); established from a pituitary tumor induced in a female Wistar/Furth rat in 1965; described in the literature to produce a high rate of prolactin and a low rate of growth hormone synthesis
Biosafety level:
1
Permissions and restrictions:
DSMZ Cell Culture Data:
Morphology:
Medium:
80-90% Ham's F10 + 10-20% h.i. FBS + 2 mM L-glutamine (originally grown with 15% horse serum + 2.5% h.i. FBS in Ham´s F10)
Subculture:
cells grow slowly and do not reach high cell densities; seed out at ca. 0.5 x 106 cells/ml; maintain at 0.3-0.8 x 106 cells/ml; adherent cells might be detached by "washing" or with TNE (40 mM TrisCl pH 7.5, 1 mM EDTA pH 7.5, 150 mM NaCl);
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 60-80 hours
Harvest:
maximal density of 0.8 x 106 cells/ml
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
DSMZ Scientific Data:
Mycoplasma:
contamination was eliminated with enrofloxacin (Baytril), then negative in microbiological culture, RNA hybridization, PCR assays
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Rattus norvegicus species.
Cytogenetics:
rat karyotype
Viruses:
PCR: SMRV -
