seed out 0.2-0.5 x 106 cells in 80 cm2; split confluent culture 1:2 once a week using trypsin-like enzyme (e.g. Tryple Express) (standard trypsin/EDTA is not sufficiently effective); note that viability may decrease rapidly after reaching confluency
