80-90% RPMI 1640 + 10-20% h.i. FBS or 80-90% MEM (with Earle's salts) + 10-20% h.i. FBS + non-essential amino acids + 2 mM L-glutamine
Subculture:
seed out at ca. 1 x 106 cells/25 cm2 flask; split confluent culture 1:2 every 3-4 days by tapping the flask or by pipetting (trypsin/EDTA is usually not necessary); viability can decrease shortly to 50-70% after thawing
Incubation:
at 37 °C with 5% CO2

ca. 100 hours

cell harvest of ca. 25 x 106 cells/175 cm2
