Subculture:
seed out at ca. 1 x 106 cells/25 cm2 flask; split confluent culture 1:2 to 1:3 every 4-6 days using trypsin/EDTA (cells are only slightly adherent and can also be detached by tapping the flask or pipetting, but trypsin/EDTA is necessary to prepare single cells); viability may drop to 60% after thawing, but cells recover within the first 3 days

at 37 °C with 5% CO2
