Subculture:
cells are difficult to culture; seed out at ca. 0.5 x 106 cells/ml, maintain at 0.4-1.0 x 106 cells/ml (do not exceed these limits), split saturated culture 1:2 every 4-5 days; upon thawing viability drop to about 50% and may need one week to recover; accumulation of
calcium phosphate (hydroxyapatite) in the culture was described in ref Harasawa and observed by us - particles can be tiny (resembling bacterial infection) or form larger, dark aggregates with irregular surface; washing of cells with PBS is recommended to
reduce the amount of debris; cells were described to be constitutively cytokine-dependent, however, we found that the cells are no more cytokine dependent.
