semi-adherent, granular collagen-rich cells of irregular shape
Medium:
80% mixture of Iscove's MDM and RPMI 1640 (at 4:1) + 20% FBS (do not heat-inactivate!)
Subculture:
seed out at ca. 7 x 106 cells/80 cm2 flask; split confluent culture 1:2; use trypsin/EDTA for subculture, although cells detach easily with trypsin/EDTA; trypsinized cells adhere evenly to the bottom of the vessel, whereas non-trypsinized cells grow as clumps in suspension; as the cells are quite loosely attached, the culture vessels should be handled cautiously when spent medium is exchanged
