Origin:
Genetic modification of human osteosarcoma cell line U-2-OS for studying sister chromatid recombination (SCR). The cell line is described to contain a single copy of the SCR reporter inserted as nested intron and carries the GFP-Gene and is puromycin resistant. A series of modified cell lines with gene disruptions in RAD51 paralogs are also available (DSMZ ACC 833-ACC 842).

Medium:
90% DMEM + 10% h.i. FBS + (additionally) 2 mM L-glutamine; (to verify puromycin resistance, 1 µg/ml should be added)
Subculture:
seed out at 1 x 106 cells/80 cm2;
split confluent culture once or twice a week 1:3 to 1:10 using trypsin/EDTA; apparently confluent cultures can be further cultivated for 2-3 days to increase the maximal cell harvest
Incubation:
at 37 °C with 10% CO2
Doubling time:
ca. 50-60 hours
Harvest:
cell harvest of 5-10 x 106 cells/80 cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
