Risk assessment:
the cell line contains parts of the hepatitis B virus (HBV) genome integrated into the eukaryotic genome. The HBV surface antigen (HBsAg) is expressed, but no active viruses are produced by the cells. The cells can thus be handled under biosafety level 1. According to the German Central Committee for Biological Safety (ZKBS) the cells can also be handled under biosafety level 1 when the cells are used for genetic modifications.

Subculture:
seed out at ca. 0.5-1 x 106 cells/80 cm2;
split confluent culture 1:4 to 1:10 every 3-5 days using trypsin/EDTA

at 37 °C with 5% CO2

about 40-50 hours

Mycoplasma:
initial contamination was eliminated with Mycoplasma Removal Agent, then negative in DAPI, microbiological culture, RNA hybridization, and continuously in PCR assays

STR analysis according to the global standard ANSI/ATCC ASN-0002.1-2021 (2021) resulted in an authentic STR profile of the reference STR database.
