The naturally occurring transposon Tn1721 (11.4 kb) was inserted in vivo into the conjugative Kmr ts plasmid Rts1 (Rts1::Tn1721); Tn1721 itself belongs to the Tn3 family and consists of a repetitious tet region and a "minor transposon" (Tn1722), the latter encoding transposition functions; useful for mutagenesis in Escherichia coli; the large size of Rts1 (217 kb) prevents cotransformation of any Rts1::Tn replicons.
