The transposon Tn1731 (7.8 kb) was inserted in vivo into the conjugative Kmr ts plasmid Rts1 (Rts1::Tn1731); Tn1731 itself is a Tcr variant of Tn1722, an in vitro recombinant of Tn1722 and a 2.4 kb EcoRI fragment carrying the tet genes of Tn1721; the EcoRI fragment was derived from plasmid pJOE452; useful for mutagenesis in Escherichia coli; the large size of Rts1 (217 kb) prevents cotransformation of any Rts1::Tn replicons.
