<- NCIMB <- M. Green, Univ. Sussex, Brighton. [WP2]. Tryptophan-requiring, radiation resistant derivative of strain B (the strain used by Luria and Delbruck in their original demonstration of mutation in bacteria). WP2 is the repair-proficient parent strain of WP2uvrA which lacks excision repair (DSM 9495). Test strain of a set for mutagen screening: in these strains, agents causing base substitution mutations can be shown to increase the frequency of Trp+ revertants. Four tests are described: Spot tests, treat and plate tests, a simplified fluctuation test and use of a liver microsomal fraction. The Trp+ reversion system does not detect agents which cause frameshift mutations. This E. coli test system is complementary to the S. typhimurium system of Ames. (Medium 381, 37°C).
