Peptidoglycan of Gram-positive bacteria for reference purposes is prepared as described in Protocol 7 (Schumann, 2011). The yield of peptidoglycan obtained from 4 g wet biomass is usually approximately 10-15 mg but cannot be guaranteed. The minimum amount provided is 5 mg. The production of amounts > 20 mg is beyond the capacities of the DSMZ Identification Service. A thin-layer chromatogram of the peptidoglycan hydrolysate serves for quality evaluation.
Required material: 3-4 g wet biomass (centrifugal pellet) sent either in frozen state on dry ice or, alternatively, suspended in isopropanol/water (1:1, v/v) at ambient temperature.
Important: Biomass must be harvested before sporulation or autolysis. Lyophilized cells are suboptimal but can also be sent in the amount corresponding to 4 g wet weight.
