85% DMEM + 15% h.i. FBS + 4 mM L-glutamine + 1 mM sodium pyruvate + 0.4 µg/ml hydrocortisone + 10 ng/ml EGF
Subculture:
split nearly confluent culture 1:2 to 1:3 about once a week using trypsin/EDTA (it may be difficult to detach the cells from the flask - use double quantity of trypsin/EDTA for ca. 15 min); seed out at ca. 2 x 106 cells/75 cm2
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 72 hours
Harvest:
cell harvest of ca. 8 x 106 cells/75 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO

initial contamination was eliminated with Baytril (enrofloxacin), then negative in DAPI, microbiological culture, RNA hybridization and continuously in PCR assays
