Origin:
Genetical modification of U2OS#18 (DSMZ ACC 832, a reporter cell line for measurement of sister chromatid recombination, SCR) with a genetic disruption of XRCC2 by frameshifting mutations in exon 3 (targeted by
CRISPR-CAS9; puromycin resistant and expressing GFP). The wild-type allele is no longer present. A sister cell line with the retrovirally reconstituted XRCC2 gene is available as ACC 841

Medium:
80% DMEM + 20% h.i. FBS + (additionally) 2 mM L-glutamine (to verify puromycin resistance, 1 µg/ml should be added)
Subculture:
seed out at 1 x 106 cells/80 cm2;
split confluent culture once a week 1:2 to 1:3 using trypsin/EDTA (wash twice with PBS before adding trypsin/EDTA); apparently confluent cultures can be further cultivated for 4-6 days to increase the maximal cell harvest
Incubation:
at 37 °C with 10% CO2

ca. 4-6 days

cell harvest of ca. 4-8 x 106 cells/80 cm2
