Medium:
90% RPMI 1640 + 10% h.i. FBS + 2 mM L-glutamine + non-essential amino acids + 1 mM sodium pyruvate + 10 µg/ml human insulin
Subculture:
seed out at ca. 0.5 x 106 cells/ml; maintain at 0.3-1.0 x 106 cells/ml; split ratio 1:3 to 1:5; avoid lipopolysaccharide contamination which will result in cell clumping, maturation and retarded growth; cells are best started in 12- or 24-well plates
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 50-60 hours
Harvest:
maximal density at ca. 1.0 x 106 cells/ml
