Origin:
established from peripheral blood lymphocytes of a cotton-top tamarin (cotton-top marmoset) monkey (Saguinus oedipus); cells were described in the literature to release high titers of Epstein-Barr virus (EBV) providing a source for transformation of B cells and establishment of continuous cell lines

Risk assessment:
The cell line is positive for EBV (HHV-4) by PCR analysis. Additionally, expression of immediate-early protein BZLF-1 and lately expressed capsid protein were also positive by western blot and immunostaining, respectively, in untreated and phorbol ester / sodium butyrate stimulated cells. The infection was classified as lytic associated with a production of active viruses. A transmission of EBV during handling of the cells is possible and the cell line is thus categorized biosafety level 2.

Morphology:
single, round to oval cells or clustered in small clumps in suspension and partially adherent; ca. 10% adhere, but trypsin is not necessary to detach the cells; image;
Medium:
90% RPMI 1640 + 10% h.i. FBS
Subculture:
maintain at 0.5-1.0 x 106 cells/ml; split 1:2 to 1:5 every 72 hours; minimal density at 0.2 x 106 cells/ml; adherent cells might be detached by tapping the flask; seed out at ca. 0.5 x 106 cells/ml
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 41 hours
Harvest:
maximal density at about 3 x 106 cells/ml

Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Saguinus oedipus species.

near-diploid simian karyotype with 4% polyploidy - 44(42-47)<2n> - chromosome rearrangements present
