ACC 192
Cell line:
EBL
DSMZ no.:
ACC 192
Species:
bovine (Bos taurus)
Cell type:
embryonic lung cells
Origin:
established from lung of a 7-month-old bovine fetus; useful for detection of Pasteurella multocida toxine
Biosafety level:
1
Permissions and restrictions:
DSMZ Cell Culture Data:
Morphology:
Medium:
90% RPMI 1640 + 10% h.i. FBS
Subculture:
seed out at about 1 x 106 cells/175 cm2; split confluent culture every 3-4 days using trypsin/EDTA; for the toxine test cells should be starved and split once a week
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 70 hours
Harvest:
cell harvest of about 15 x 106 cells/175 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
DSMZ Scientific Data:
Mycoplasma:
originally negative in DAPI, microbiological culture, RNA hybridization assay, and recently by PCR
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Bos taurus species.
Cytogenetics:
apparently bovine karyotype with bimodal ploidy distribution - 42(40-42)<2n>/80(77-84)<4n> - structurally rearranged with apparent reduction of ch number by centric fusion followed by tetraploidization
Viruses:
ELISA: reverse transcriptase positive; PCR: SMRV -, BVDV +
