Origin:
IL-3 dependent murine pro B cell line; an original publication describing establishment of BA/F3 does not exist; probably a subclone of a first described Ea3 cell line. Performing May-Gruenwald-Giemsa staining provides evidence that the cells are indeed from a lymphatic background, however cells were apparently not derived from BALB/c, but from C3H mouse. 95% RPMI 1640 + 5-% h.i. FBS + 10 ng/ml mouse IL-3 (or 10% vol conditioned medium of WEHI-3B cell line, DSMZ ACC 26)
Subculture:
seed out at about 1-3 x 105 cells/ml; split saturated culture 1:10 every 3 days; at high cell density (>2 x 106 cells/ml), a cytokine-independent subclone may grow out relatively quickly. CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus, the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines.
