established from the tumor of a 60-year-old man with moderately differentiated squamous cell carcinoma of the soft palate which was invasive into muscle layer; cell line was established from a cervical lymph node metastasis which occurred seven years after initial treatment of the primary tumor; the tumor metastasized not only to the lungs, but also to the brain; described as anchorage-independent and tumorigenic in nude mice

Medium:

90% DMEM (4.5 g/L glucose) + 10-20% h.i. FBS, 20% h.i. FBS in the first week

Subculture:

split confluent culture 1:2 - 1:4 every 7 days using trypsin/EDTA; seed out at ca. 1.5 x 10⁶ cells/80 cm² in 15 ml medium

Incubation:

at 37 °C with 10% CO₂

Doubling time:

ca. 100 hours

Harvest:

cell harvest of ca. 8-10 x 10⁶ cells/175 cm²

Storage:

frozen with 70% medium, 20% FBS, 10% DMSO
