95% Iscove's MDM + 5% h.i. FBS + 2 mM L-glutamine + 2.5 x 10-5 M mercaptoethanol + IL-3 (either 30% vol conditioned medium of WEHI-3B cell line (DSM ACC 26) or 20 U/ml mIL-3)
Subculture:
maintain at 0.2-0.4 x 106 cells/ml; split 1:2 to 1:3 every 1-2 days, cells grow fast. CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines.

at 37 °C with 5% CO2

maximal density at 0.8 x 106 cells/ml

frozen with 70% medium, 20% FBS, 10% DMSO
