the cells are cultivated serum-free in NS-A medium + 20 ng/ml EGF + 20 ng/ml FGF2 + 10 µg/ml Heparin, modified SOP CELL307-NSA but without antibiotics
Subculture:
cells are difficult to culture; cultivate in low-adherent 6-well plates; seed out all cells (approx 2 x 106 cells) into 1 well with about 4 ml medium; change/add medium when turning into orange-yellow or at least once a week; disperse aggregates gently 1-2 times a week and only when a size of about 0.5-1 mm is reached (use pipettes with wide opening (serological, wide-bore or cut); optimal density 6-10 large spheroids/well. Culture plates should not be used longer than 4 weeks

at 37 °C with 5% CO2

ca. 8-10 days
Harvest:
6-10 large spheroids in one well of a 6-well may contain up to 2 x 106 cells
