Polynucleobacter strains grow so weakly that single colonies or a streak of colonies might not be visible on the agar surface. They grow reliably only if the agar surface is moist. We grow the cultures generally on agar slants with 1.5 -2 ml liquid media added. The slants are inoculated and incubated upright for about 1 week. The bacteria will grow and settle down as a faint white precipitate at the ground of the slant (deepest point of liquid). That is how the cultures are prepared for delivery also. Transfer about 250 µl of the suspension to inoculate fresh slants. Growth will be indicated by the precipitate.
